Metabarcoding analysis and fermentation performance of the dominant fungal microbiota associated with the Algerian traditional date product “Btana”

The study highlights the fungal diversity of traditional Algerian date’s product “Btana” prepared with direct Btana method (DBM) and undirect (UBM). populations were analyzed through 28S metabarcoding. Data treatment resulted in 122,997 reads representing three Phyla which 76% (46 OTUs) belong to Ascomycota phylum. Zygosaccharomyces rouxii was most prevailed species accounting for 35.40% total population. Similarity percentage analysis revealed a low level resemblance each two types (DBM: 17.26%, UBM: 16.87). According HPLC analysis, lactate detected nine samples within range 0.87-23.06 g/100g. Culture plating subsequent D1/D2 domain 28s DNA showed prevalence Z. rouxii. Fermentation non-renewed date medium high ethanol production (21.31 ± 2.89 g/100g) by Lachancea thermotolerans 5.87 g/100g lactates Kluyveromyces delphensis. Enzymatic assay esterase (C4) naphtol-AS-BI-phosphohydrolase activity L. thermotolerans, K. delphensis, Pichia subpelliculosa, while ?-fucosidase recorded P. subpelliculosa. current results demonstrated that is promising source yeasts useful value-added products like bioethanol lactic acid using low-income cultivars.
